Undergraduate Thesis (BSE major in Biology) Pamantasan ng Lungsod ng Maynila, 2004.;ABSTRACT : This study was conducted to determine the fungicidal potential of Camachile (Pithecolobium dulce) bark extract against the Black mold (Aspergillus niger). The barks of the Camachile tree were extracted using water as the solvent medium. Preparations of the ratio 1:2 or 50% extract concentration were administered to the test organism. Sabouraud's medium were used in preparing/culturing the black mold. For the purpose of comparison, a commercially available fungicide were utilized as the positive treatment. Assay plate method were used to test the anti-fungal potential. Ten samples were used for both the extract and the positive treatment. The effectiveness of the extract were indicated by the zone of inhibition - the clear zone around the filter paper disc prepared - measured in millimeter. For conformity purposes, another test were employed using Agar Incorporation Assay at 1ml/plate, another 1 ml of commercially available fungicide were mixed also in 10 ml Agar to determine if the cultured organism will grow. Results showed that there was significant difference between the zones of inhibition of black mold produced by Camachile bark extract produced negative inhibition while commercially available fungicide revealed positive inhibitory action against the test organism. This is attributed to the solvent medium and the method of extraction that were used which affected the concentration of the extract. Findings also showed that the test organism's growth were inhibited using the Agar Incorporated Assay wherein the extract were directly inoculated to the Agar Plate. Observations on the morphology of the black mold seen under the low power objective treated and not treated with the extract showed that there were no change in black mold's mycelium, spore - bearing head and color. 56
