ABSTRACT: The study aimed to investigate the cytogenetic and mutagenetic effect of water samples collected from Pasig River in terms of root length, forms, colors, turgescences, mitotic indexes, c-mitosis, chromosome stickiness and bridges. This study employed the experimental research design. To assess the water from Pasig River using Allium test, water samples were collected from the two stations of the river. The samples have undergone heavy metal analysis. The study followed modified form of the test where onions were directly germinated in the collected water samples. The cytogenetic and mutagenetic effects of the water samples were computed and analyzed statistically to determine the relationship of the mean statistically. The extent to which the mean scores were related was analyzed using one-way ANOVA and Turkey's Honest Significant Difference was applied to show the significant differences of the mean scores. The study revealed that there were significant differences on the cytotoxic effect of heavy metals exhibited using Allium test with regards to the variables among the sampling stations and there were significant differences on the mutagenetic effect of heavy metals exhibited using Allium test with regards to the variables among the sampling stations. A laboratory activity was made from assessment of water samples from Pasig River using Allium test which can be utilized in High School Genetics. The laboratory activity focused on mitotic cell division and computation of mitotic indices that correspond to different amounts of heavy metals present in the said river.
