ABSTRACT: This study focuses on the biosynthesis of L-tryptophan using glycine and indole-3-acetic acid by E. coli. Using 3 different culture media I can interpret if there is significant difference between the 3 culture media that is used for the growth curve of the E. coli. Three different culture media were used for the E. Coli growth curve and at the mid-point value were used for statistical treatment and showed that there is no significant difference between the 3 culture media at its mid-point value. Biosynthesis was made at the mid-point of the culture media using glucine and indole-3-acetic acid by E. Coli and after an hour, centrifugation was used for the lyses of the bacteria and releasing of the synthesize L-tryptophan. Statistical treatment of the growth curve of the WE. Coli at the mid point value showed that there is no significant different between the three culture media at this point. Results showed that the E. coli grown on 3 different culture media didn't synthesize L-tryptophan. The possible reason for this problem might be that the E. coli that was used has a special characteristics of having a membrane-bound enzyme into which it will not release its product in the cytoplasm or outside the cell.
